Louis Pasteur: Contributions and the Father of Microbiology

Who was Louis Pasteur and why is he called the father of microbiology? His work on germ theory, fermentation, pasteurization, and the first attenuated vaccines.

On the morning of July 6, 1885, a nine-year-old boy named Joseph Meister was carried into Louis Pasteur's laboratory in Paris. Two days earlier he had been bitten fourteen times by a rabid dog, some of the wounds deep enough that he could barely walk. At the time, rabies meant a certain and horrifying death; no one bitten badly enough to develop it had ever survived.

Pasteur was not a physician and had never given his experimental rabies treatment to a human. His vaccine, made from the dried spinal cords of infected rabbits, had protected dogs, but using it on a child was another matter entirely. Under pressure, and convinced the boy would otherwise die, Pasteur agreed. Over ten days, a series of injections was given, each slightly more virulent than the last. Joseph Meister never developed rabies. He lived.

That single case, more than any paper or lecture, announced that disease could be prevented by design, in the laboratory, using weakened microbes. It is a fitting place to begin the story of the man often called the father of microbiology.

Who was Louis Pasteur?

Louis Pasteur (1822-1895) was a French chemist and microbiologist whose work helped found the science of microbiology. Trained originally as a chemist, he went on to show that microorganisms cause fermentation and disease, to disprove the old idea that life arises spontaneously from non-living matter, to invent the process now called pasteurization, and to develop the first vaccines made from weakened microbes.

He is often called the father of microbiology because his work established, more clearly than anyone before him, that microscopic living organisms shape the visible world: they spoil wine, ferment beer, cause disease, and can be harnessed to prevent it. He shares the broad founding of the field with Robert Koch, who is more specifically called the father of bacteriology.

Fermentation: microbes do the work

Pasteur's path into microbiology began not with disease but with industry. In the late 1850s, working in the wine and beer regions of France, he was asked why some batches of wine and beer spoiled. The accepted view was that fermentation was a purely chemical process.

Pasteur showed instead that fermentation is carried out by living microorganisms, and that spoilage happens when the wrong microbes take over. Good fermentation and bad spoilage were both microbial, just different organisms. This was the first time anyone had firmly tied a major industrial process to the activity of specific microbes.

The discovery founded the study of microbial fermentation, which underlies the making of bread, cheese, yogurt, wine, and beer, and the modern use of defined starter cultures to control it.

Disproving spontaneous generation

If microbes cause fermentation and spoilage, where do the microbes come from? The long-held answer was spontaneous generation: the belief that living things could arise on their own from non-living matter, that maggots simply appeared in meat and microbes simply appeared in broth.

Pasteur ended the debate with an elegant experiment. He boiled broth in flasks with long, S-shaped "swan necks." The bend let air reach the broth freely but trapped dust and airborne microbes on the way down. The broth stayed sterile for as long as the neck was intact. Tilt the flask so the broth touched the trapped dust, or break the neck, and the broth quickly teemed with life.

The conclusion was decisive: microbes in the broth came from other microbes carried in the air, not from the broth itself. Life comes from life. This cleared the way for the germ theory of disease, because it established that microbes are specific organisms with a source, not spontaneous appearances.

Germ theory of disease

Pasteur's fermentation and spontaneous-generation work led him to a larger idea: if microbes spoil wine, perhaps microbes also cause disease. This is the germ theory of disease, the principle that many diseases are caused by specific microorganisms.

Pasteur did not invent the idea from nothing; others had suspected it. But his experiments gave it a solid experimental footing, and he, along with Robert Koch, is credited with establishing it as the foundation of modern medicine. Pasteurization

While studying wine and beer spoilage, Pasteur found a practical solution that carries his name to this day. He showed that gently heating a liquid to a specific temperature for a set time killed most of the spoilage microbes without ruining the taste. This process is now called pasteurization, and it is used worldwide to make milk, juice, and other products safe.

Vaccination: the first attenuated vaccines

Pasteur's most famous contribution is to vaccination, and it is worth being precise about what was and was not new. Edward Jenner had introduced smallpox vaccination decades earlier, in 1796, using cowpox. Pasteur's advance was different: he found a general method to weaken (attenuate) a disease-causing microbe in the laboratory and use the weakened form as a vaccine. He also coined the word "vaccine" in Jenner's honor, extending it from smallpox to any such preparation.

His vaccine work came in three steps:

  • Fowl cholera (1879): the discovery was an accident. Cultures of the fowl cholera bacterium, left standing over a holiday, lost their power to kill chickens. Birds injected with the old, weakened culture survived and then resisted fresh, virulent bacteria. Pasteur recognized the accident as a method: a microbe could be weakened yet still trigger protection.
  • Anthrax (1881): Pasteur applied the same principle to anthrax. In a famous public demonstration at Pouilly-le-Fort in 1881, his team vaccinated farm animals with weakened Bacillus anthracis, then exposed both vaccinated and unvaccinated animals to virulent anthrax.   • Rabies (1885): the rabies vaccine, made from dried spinal cords of infected rabbits, was Pasteur's boldest step, because rabies was untreatable and the first human patient was a child. The Pasteur Institute

The flood of rabies patients who came to Paris after Meister's cure led to the founding of the Pasteur Institute in 1887, inaugurated in 1888, dedicated to research on infectious disease and to vaccine production. Pasteur led it until his death in 1895. It remains one of the world's leading infectious-disease research centers and was, a century later, among the first to isolate HIV.

Why Louis Pasteur is called the father of microbiology

Pasteur earns the title "father of microbiology" because his work touched nearly every root of the field at once: he proved microbes drive fermentation, showed they do not arise spontaneously, helped establish that they cause disease, invented a way to make food safe from them, and found a general method to turn them into vaccines.

He is usually paired with Robert Koch, and the two are best kept straight by what each is most credited with.

Where Students Get Confused

Koch versus Pasteur. Both are founding fathers, but of different things. Pasteur is the father of microbiology in the broad sense: fermentation, germ theory, spontaneous generation, vaccines. Koch is the father of bacteriology and medical microbiology: proving specific bacteria cause specific diseases, and the pure-culture methods and postulates to do it.

Pasteur did not make the first vaccine. Edward Jenner did, against smallpox, in 1796. Pasteur's contribution was the general method of attenuating microbes in the laboratory, and the first vaccines made this way (fowl cholera, anthrax, rabies). He named the concept "vaccine" in Jenner's honor.

Pasteur did not single-handedly invent germ theory. The idea existed before him. Pasteur (with Koch) gave it firm experimental proof and made it the foundation of medicine. Saying he "proposed" or "proved" it alone is an overstatement.

Pasteurization does not sterilize. It kills most spoilage and disease microbes but not all, and not spores. It makes food safe and longer-lasting, but a pasteurized product is not sterile.

Pasteur was a chemist, not a physician. He never held a medical degree. This is why the first human rabies treatment was so fraught, and why a physician administered the injections to Joseph Meister.
