Utilization tests ask whether a bacterium can use a defined compound as a nutrient or can break it down with a specific enzyme. Their media and color changes can look similar, but the tests answer different identification questions. Which utilization test should you study first?

Start with the citrate utilization test. It introduces the core pattern: a defined carbon source, growth on a minimal medium, alkaline products, and a bromothymol-blue color change. Citrate is commonly used within enteric identification panels.

Next, compare the acetate utilization test. Acetate agar uses sodium acetate as the sole carbon source and helps separate most Escherichia coli strains from most Shigella strains. The article explains important exceptions, so the result should never be interpreted alone.

How is acetamide different from acetate?

Acetate, by contrast, is a carbon-utilization test used in an enteric differentiation panel. Similar names and the same green-to-blue indicator make this a frequent source of confusion.

What should you compare across these tests?

For every utilization test, record the substrate, the basal medium, the source of carbon and nitrogen, the indicator, the positive-control and negative-control organisms, the incubation conditions, and the exact definition of a positive result. Note whether growth alone is significant or whether a color change is also required. Finally, document exceptions and combine the result with the rest of the biochemical identification panel.
